A major challenge in obesity treatment is weight rebound after successful interventions, whether through pharmacological treatments (e.g., GLP-1 receptor agonists such as Wegovy or Ozempic) or lifestyle approaches like diet and exercise. This “yo-yo effect” is poorly understood at the cellular and molecular level.
The Chun-Xia Yi Lab received a ZonMw Vici grant to study how microglia–neuron interactions in the hypothalamus contribute to obesity and post-weight-loss weight regain. The project has two complementary branches:

Our overarching goal is to understand how the brain regulates body weight. A central focus is using human brain tissue to address fundamental questions about human brain function. With the Netherlands Brain Bank, founded by Prof. Dick Swaab, we study brains from individuals with obesity and/or diabetes to identify novel, druggable targets. These targets are investigated in animal models to uncover cellular mechanisms and evaluate potential therapies. In parallel, we conduct clinical studies in patients with metabolic disorders using non-invasive imaging to translate findings toward clinical applications.
Amsterdam UMC Research BV
Amsterdam UMC Research BV supports non-profit scientific research. In doing so, we provide researchers with everything they need to excel. Our principal investigators (PIs) and project leaders offer support in the field of project management, finance and human resources. In medical scientific research projects, legal support is also provided.
